In 1981, Plenum Press published a text entitled The Nature and Treatment of the Stress Response by Robert Rosenfeld, M. D. , and me. That text attempted to do what no other text from a major publisher had previously attempted, that is, to create a clinically practical guide for the treatment of excessive stress and its arousal-related syndromes-this to be captured between the same covers in combination with a detailed, clinically relevant pedagogy on the neurological and endocrinological foundations of the stress re sponse itself. Endoplasmic reticulum (ER) is an intracellular organelle responsible for protein folding and assembly, lipid and sterol biosynthesis, and calcium storage. A number of biochemical, physiological, or pathological stimuli can interrupt protein folding process, causing accumulation of unfolded or misfolded proteins in the ER lumen, a condition called “ER stress”. To cope with accumulation of unfolded or misfolded proteins, the ER has evolved a group of signaling pathways termed “Unfolded Protein Response (UPR)” or “ER stress response” to align cellular physiology. To maintain ER homeostasis, transcriptional regulation mediated through multiple UPR branches is orchestrated to increase ER folding capacity, reduce ER workload, and promote degradation of misfolded proteins. In recent years, accumulating evidence suggests that ER stress-triggered transcriptional reprogramming exists in many pathophysiological processes and plays fundamental roles in the initiation and progression of a variety of diseases, such as metabolic disease, cardiovascular disease, neurodegenerative disease, and cancer. The cell cycle is a sequence of biochemical events that are controlled by complex but robust molecular machinery. This enables cells to achieve accurate self-reproduction under a broad range of conditions. Environmental changes are transmitted by molecular signaling networks, which coordinate their actions with the cell cycle. This work presents the first description of two complementary computational models describing the influence of osmotic stress on the entire cell cycle of S. cerevisiae. Our models condense a vast amount of experimental evidence on the interaction of the cell cycle network components with the osmotic stress pathway. Importantly, it is only by considering the entire cell cycle that we are able to make a series of novel predictions which emerge from the coupling between the molecular components of different cell cycle phases. The model-based predictions are supported by experiments in S. cerevisiae and, moreover, have recently been observed in other eukaryotes. Furthermore our models reveal the mechanisms that emerge as a result of the interaction between the cell cycle and stress response networks.
